Description
The Sulforhodamine B (SRB) cytotoxicity assay is a sensitive, reproducible and easy-to-use assay based on the ability of SRB to bind to cellular protein components that have been immobilized on tissue culture plates.

SRB is a bright pink amino x-ton dye with two sulfonic acid groups. They bind to basic amino acid residues under mild acidic conditions and dissociate under alkaline conditions.

Since the binding of SRB is stoichiometric, the amount of dye extracted from stained cells is directly proportional to the cell mass. The fixed dye is dissolved, and a 690 nm reference filter is used for photometric measurement at OD 540 nm. The OD value is related to the total protein content and therefore to the number of cells.
