Fc Engineering Service for Fc Silence

In therapy, the Fc region of antibody provides useful properties for its use, such as a long circulating half-life and good biochemical and biophysical stability. More importantly, the Fc region interacts with various Fcγ receptors (FcγR) and complement proteins and mediates immune effector functions, such as elimination of target cells through antibody-dependent cytotoxicity (ADCC), phagocytosis (ADCP), or complement-dependent cytotoxicity (CDC).

However, there have more benefits to have a silent Fc for therapeutic approaches. Eliminating the interaction between Fc/FcγR and complement C1q can prevent unnecessary side effects, such as cell and tissue damage caused by FcγR mediated immune effect functions. Therefore, the development of a completely silenced Fc component is of great value for the development of therapeutic molecules that do not require the function of FcγR or C1q mediated effector function.

(A) Point mutations in the Fc to disrupt antibody effector functions.

(B) Exchanging amino acids between two Fc molecules from different IgG subclasses to disrupt effector functions.

(C) The removal of Asn297 glycan in the IgG Fc severely reduces the Fc binding to FcγRs by inducing an Fc closed conformation.
