Description
Fibrin is a group of proteins that contain both a collagen-like domain and a fibrinogen-like domain. Three forms of fibrin have been identified in humans: Ficolin-1 (M-ficolin), Ficolin-2 (L-ficolin) and Ficolin-3 (H-ficolin). Ficolin-2 is mainly produced by the liver. Ficolin-3 is produced by the liver and lungs, while Ficolin-1 is produced by monocytes, white blood cells and lung cells. Ficolin-2 can recognize different danger-related molecular patterns (DAMP), such as the GlcNAc structure in lipoteichoic acid (LTA) and fungal 1,3-β-D-glucan. Ficolin-2 also recognizes N-acetylated compounds, such as acetylcholine. In addition, Ficolin-2 recognizes apoptotic cells and participates in the removal of host cells. Ficolin-2 combines with MASP-2 to activate the lectin complement pathway. According to reports, Ficolin-2 serum levels have different concentrations. The polymorphism in the FCN2 gene promoter derived from Ficolin-2 is related to the change of Ficolin-2 serum level. These polymorphisms may affect the affinity of Ficolin-2 to the structure of GlcNAc. Ficolin-2 deficiency has not been reported, but low serum levels of Ficolin-2 are related to repeated respiratory infections in children. Interestingly, Ficolin-2 is associated with unique immune challenges during pregnancy. Compared with normal non-pregnant women, the content of Ficolin-2 in the plasma of normal pregnant women increased by 4 to 5 times. However, the significantly reduced Ficolin-2 plasma concentration in pregnant women is associated with pregnant women with preeclampsia. Therefore, it is necessary to evaluate Ficolin-2 to study its regulatory role in the innate immune system. The concentration of Ficolin-2 in normal human plasma ranges from 1 to 14 μg/ml.
Applications
The human Ficolin-2 ELISA kit will be used for in vitro quantitative determination of human Ficolin-2 in serum, plasma and cell culture supernatant samples.
Principle
Human Ficolin-2 ELISA kit is a ready-to-use solid-phase enzyme-linked immunosorbent assay based on the sandwich principle. The working time is 3 hours. The effective format of two plates with twelve disposable 8-well strips allows free choice of batch size for determination. Samples and standards are captured by specific antibodies bound to the solid phase. The biotinylated tracer antibody will bind to the captured Ficolin-2. The streptavidin-peroxidase conjugate will bind the biotinylated tracer antibody. The streptavidin-peroxidase conjugate will react with the substrate tetramethylbenzidine (TMB). The enzyme reaction is stopped by adding oxalic acid. Measure the absorbance at 450nm with a spectrophotometer. Draw a standard curve by plotting the absorbance (linear) relative to the corresponding concentration (logarithm) of the Ficolin-2 standard. The concentration of the human Ficolin-2 sample running at the same time as the standard can be determined from the standard curve.

Specimen Collection
Blood samples were collected using sterile venipuncture techniques, and serum was obtained using standard procedures. At least 5 ml of whole blood is recommended. Let the blood clot in the serum test tube for 60-65 minutes at room temperature (20-25°C). Centrifuge the blood sample and transfer the cell-free serum to a clean test tube. The serum must be handled correctly to prevent complement activation in vitro. Serum should be frozen in a sealed tube at -70°C or lower for long-term storage or transport on dry ice. The sample should not be frozen and thawed more than once. Do not use jaundice, lipemia and hemolytic serum. You cannot use heat-inactivated serum. Plasma cannot be used.
