The immense diversity and complexity of glycan structures and their multiple modes of interaction with proteins pose great challenges to the development of therapeutic antibodies. Various tools for glycan profiling based on chromatography, mass spectrometry, glycan microarray and glycoinformatics are being developed. Lectins, which have long been used in glycoimmunology, are printed on microarrays, providing a universal platform for rapid high-throughput analysis of glycoforms in biological samples. Lectin microarray technology surpasses traditional mass spectrometry technology for glycomic analysis in both efficiency and function. We selected 26 lectins to be included in our first-generation lectin microarray. These 26 lectins represent most of the epitopes that bind to carbohydrates and have been characterized on our glycan array. There are 8 or 16 identical sub-arrays on a single array chip, so 8 or 16 samples can be analyzed simultaneously. Our lectin microarrays provide scientists with powerful and sensitive tools for analyzing the glycosylation profiles of therapeutic proteins, biomarkers or other proteins of interest.
Lectin microarrays have been prepared using multiple immobilization techniques (1) using lectins derived from multiple sources (2). The binding of glycoconjugates (3) of analytes from multiple biological samples has been tested using multiple detection methods (4). Lectin microarrays provide fast, sensitive and high-throughput screening tools for sugar profile analysis, which can be used for important biomedical applications (5). The simultaneous application of lectin microarray and complementary glycomics technology has the potential to decode the "glycosyl code" and develop new effective diagnostic and therapeutic methods.
