The complement system plays an important role in chronic, autoimmune and infectious diseases. It contains a set of proteins that, when activated, cause the lysis of target cells and promote phagocytosis through opsonization. This plasma protein system can be directly activated by pathogens or indirectly by pathogen-bound antibodies, leading to a series of reactions involving more than 35 proteins that occur on the surface of pathogens and producing active ingredients with multiple effector functions.
There are three pathways for complement activation, namely the classic pathway, the alternative pathway and the recently discovered MB-lectin pathway. Creative Biolabs provides a complete solution for complement function analysis. Our supplement product portfolio includes active biomarkers and functional assessment of all three pathways. These assays are designed as flexible and easy-to-use enzyme immunoassays and off-the-shelf reagents, which provide a short incubation time, thereby shortening the user's hands-on time.
With our extensive knowledge of complement function testing, our in-house laboratory services have been developed to comply with GLP, GCP and ISO 17025, ensuring compliance with regulatory requirements. A comprehensive menu of markers for complement function and activation is provided, as well as a test package for evaluating complement defects and complement function.
Creative Biolabs provides complement function assays for various applications in drug development and clinical research:
Our complement function is determined by ELISA, which can be carried out on higher through-hole 96-well plates, and depends on the deposition of complement components on the activated surface of the plate. Our complement function/activity test includes:
Sometimes, it is important to check the function of individual complement components to investigate suspected complement deficiency or evaluate its inhibitory effects. The total function determination depends on all the components and functions present, and due to the relative abundance of the components, the component you are interested in may not be the rate-limiting component. Studying the functions of individual components can be very helpful. Therefore, Creative Biolabs is about to conduct separate hemolysis analysis for C2, C3, C4 and C5 and C1, B factor, D, C6, C7, C8 and C9.
With our extensive knowledge of complement function testing, we combine our experience with testing to activate or inhibit complement in vitro. By exposing normal human or NHP serum with the test article of interest, we can measure the level of complement activation or complement inhibition. For this test, we have established a custom protocol, so inquire and we can customize a plan for you.
