ADCP is mediated by monocytes, macrophages, neutrophils, and dendritic cells to engulf disease cells by expressing FcγRIIa (CD32a), FcγRI (CD64), and FcγRIIIa (CD16a), and blockade studies have shown that FcγRIIa is the primary FcγR receptor involved in this process. Complement is a group of enzymatically active proteins found in human or vertebrate serum and tissue fluids, including more than 30 soluble and membrane-bound proteins. The antibody first binds to complement C1q, and then C2-C9 is activated to form an attack complex that exerts a lytic effect on the target cells. Among the four common IgG isoforms, IgG1 and IgG3 can induce strong ADCC effects, but IgG1 has a longer half-life and a more stable structure than IgG3, so ADCC effector antibodies usually choose the IgG1 isoform. To exert the ADCC effect, the antibody needs to bind to the Fc receptor (FcγR). Antibodies typically bind to FcγR through the hinge region and the CH2 region. The Asn-N297-linked glycosylation of the CH2 domain and the modified glycoform components (such as acetylglucosamine, mannose, fucose, and sialic acid) determine the stability and binding properties of CH2 structure. Based on the important role of ADCC in the research and development of antibody drugs, it is hoped that the activation effect of ADCC can be improved by changing the design of antibodies, and the main way is to improve the binding ability of Fc to FcγR, which is achieved through amino acid mutation and glycosylation modification in technical strategy. Therefore, the detection of ADCC is inseparable from the biological activity analysis and quality attribute control of therapeutic antibody drugs.
