Description
Fibrin is a group of proteins that contain both a collagen-like domain and a fibrinogen-like domain. Three forms of Feikelin have been identified in humans: Feikelin 1 (M-ficolin), Feikelin 2 (L-Ficolin) and Feikelin 3 (H-Ficolin 3). Ficolin-3 (also known as Bodo antigen or H-ficolin) consists of a collagen-like chain and three C-terminal recognition domains that bind to acetyl groups on the surface of microorganisms (such as GlcNAc or GalNAc). Ficolin-3 is synthesized in the liver (from where it is secreted into the blood circulation) and the lungs. Similar to MBL and Ficolin-2, Ficolin-3 relies on MBL-related serine protease 2 (MASP-2) to activate the complement system. After the Ficolin-3 / MASP-2 complex binds to the surface of the microorganism, MASP-2 cleaves C4 and C2 in turn, thereby producing the C3 convertase C4bC2b, which ultimately leads to opsonization, direct lysis of pathogens and recruitment of inflammatory cells. The average concentration of Ficolin-3 in serum is 15μg/ml, with only a small change. Except for some patients with systemic lupus erythematosus, Ficolin-3 is present in all serums of more than 150,000 people. It was found that about 5% of patients with systemic lupus erythematosus were Ficolin-3 negative, possibly due to the presence of anti-Ficolin-3 autoantibodies. Ficolin-3 is always present in 398 patients with other autoimmune diseases. In liver disease, serum levels decrease with increasing severity of cirrhosis. Ficolin-3 binds to late apoptotic cell populations, and at the same time a firm and uniform binding to necrotic cells is observed. The binding properties are different from MBL and Ficolin-2. The binding of Ficolin-3 to late apoptotic cells resulted in a significant increase in macrophage adhesion/uptake.
Applications

Principle
Human Ficolin-3 ELISA kit is a ready-to-use solid-phase enzyme-linked immunosorbent assay based on the sandwich principle. The working time is 3 hours. The effective format of two plates with twelve disposable 8-well strips allows free choice of batch size for determination. Samples and standards are captured by specific antibodies bound to the solid phase. The biotinylated tracer antibody will bind to the captured Ficolin-3. The streptavidin-peroxidase conjugate will bind the biotinylated tracer antibody. The streptavidin-peroxidase conjugate will react with the substrate tetramethylbenzidine (TMB). The enzyme reaction is stopped by adding oxalic acid. Measure the absorbance at 450nm with a spectrophotometer. Draw a standard curve by plotting the absorbance (linear) against the corresponding concentration (logarithm) of the Ficolin-3 standard. The concentration of the human Ficolin-3 sample that runs simultaneously with the standard can be determined from the standard curve.

Specimen Collection
Blood samples were collected using sterile venipuncture techniques, and serum was obtained using standard procedures. At least 5 ml of whole blood is recommended. Let the blood clot in the serum test tube for 60-65 minutes at room temperature (20-25°C). Centrifuge the blood sample and transfer the cell-free serum to a clean test tube. The serum must be handled correctly to prevent complement activation in vitro. Serum should be frozen in a sealed tube at -70°C or lower for long-term storage or transport on dry ice. The sample should not be frozen and thawed more than once. Do not use jaundice, lipemia and hemolytic serum. You cannot use heat-inactivated serum. Plasma cannot be used.
