Annexin V-PE Apoptosis Detection Kit Plus is based on the following observations: Most cell types transfer membrane phosphatidylserine (PS) from the inner surface of the plasma membrane to the cell surface soon after initiating apoptosis. Once it reaches the cell surface, PS can be easily detected by staining with the fluorescent conjugate of Annexin V, which is a protein with a strong natural affinity for PS. The one-step dyeing process only takes 10 minutes. In addition, the assay can be performed directly on live cells without fixation. Annexin V-PE Apoptosis Detection Kit Plus includes Annexin V-PE, SYTOX green dye and binding buffer. SYTOX green dye is impermeable to living cells and apoptotic cells, but necrotic cells have strong green fluorescence by binding cell nucleic acid. After staining the cell population with Annexin V-PE and SYTOX green dye in the provided binding buffer, apoptotic cells show red fluorescence, dead cells show green fluorescence, and live cells show little or no fluorescence. These populations are easily distinguished by fluorescence microscopy or flow cytometry using FITC and PE filters, where SYTOX Green dye uses the FL1 channel (eg 488 nm/Em. 530 nm) and Annexin V-PE uses the FL2 channel (For example, 488 nm) /Em.578 nm).

Detect the apoptosis of living cells by flow cytometry or fluorescence microscope. PE shows brighter red fluorescence. Sytox green dye is also included to control necrosis.
