Ubiquitin C-terminal hydrolase 1 (UCH-L1) is a deubiquitinating enzyme (DUB), which is mainly expressed in neuronal tissues and accounts for 1-2% of total protein in certain brain tissues. UCH-L1 is a cysteine ​​protease that can cleave the isopeptide bond between the carboxyl end of the ubiquitin molecule and the lysine residue on the ubiquitin-modified protein. Since ubiquitination can be used for both the localization and degradation of these modified proteins, the proper function of deubiquitination enzymes is essential for cell viability and integrity. The disorder of UCH-L1 is related to the pathophysiology of neurological diseases such as Parkinson's disease and Alzheimer's disease and the invasion of cancer. The identification of inhibitors with high selectivity can facilitate a further understanding of these processes. More than 400 DUB enzymes are known, and this target has higher selectivity than the proteasome system.
