Description
The monoclonal antibody 3A3E1 recognizes human properdin, also known as complement factor P. The complement system is the first line of defense against pathogens and helps eliminate apoptosis and damaged cells. Positively regulates plasma protein properdin. The alternative pathway of complement is crucial. It is a single-chain glycoprotein (ca 53k Da) composed of six TSR sequences, which exists in the blood as a mixture of preferred head-to-tail trimers, dimers and tetramers The protein produces T lymphocytes and T lymphocytes from white blood cells (such as activated neutrophils and monocytes), and can also be passed through, for example, stressed endothelial cells.
Properdin can start with C3 and factors B, D, I and H and actively regulate the activity of the alternative pathway. It binds to C3b, thereby stabilizing the unstable C3bBb convertase deposited on the immune complex or foreign surface. Thus, AP can be enhanced by stimulating the amplification of C3bBb convertase, which competes with factor I catabolism, and factor I uses factor H as a cofactor. The local amplification process leads to the production of the alternative pathway C5 convertase C3bBb3b and initiates the terminal pathway of complement activation. The alternative pathway can account for about 80% of the terminal pathway activity. Properdin has also shown the ability to directly limit the activity of factor H.
Recent studies have shown that properdin is also a pattern recognition receptor (PRR) that can directly bind to the surface of microorganisms, apoptotic and necrotic cells (dangerous non-self and altered self). Inappropriate activation or dysregulation of alternative pathways is a key factor in the development of several autoimmune diseases. Targets opsonized with properdin are marked as cleared by scavenger cells, even without complement. This makes it a potential target for disease treatment. Recent studies have shown that people have renewed interest in the role of properdin in the pathogenesis of diseases such as asthma, arthritis, septic shock, AMD and C3 glomerulopathy.
Antibody 3A3E1 can be used in ELISA, Western blotting (only non-reduction method), and as an antibody for functional research, which can inhibit alternative pathways of complement, such as standard red blood cell AP50 analysis.
