Description
Mannose binding lectin (MBL), also known as mannose or mannan binding protein (MBP), is a member of collectins and an important part of innate immunity. MBL is an oligolectin that can recognize carbohydrates as mannose and N-acetylglucosamine on pathogens. MBL contains a recognition domain rich in cysteine, similar to collagen and carbohydrates. It forms a complex with C1r/C1, such as the serine protease MASP, which cleaves C4, C2, and C3 through proteolysis. MBL can activate the complement pathway independently of the classical and alternative complement activation pathways. The MBL-MASP pathway (more called the lectin pathway) is independent of antibodies and C1q. MBL exhibits complement-dependent antibacterial activity and directly acts as a conditioning agent, so it plays an important role in innate immunity. MBL is synthesized by hepatocytes and has been isolated from the liver and serum of several vertebrates. Only one form of human MBL has been characterized, while two forms have been found in rhesus monkeys, rabbits, rats and mice. The murine forms are called MBL-A and MBL-C. The concentration of MBL-C in serum is about 6 times higher than that of MBL-A. In casein and LPS injection models, MBL-A was found to be an acute phase protein, but not MBL-C. MBL-C exists in a higher oligomeric form than MBL-A. In the plasma of healthy mice, the concentration of MBL-A ranges from 4 to 12 μg/ml. In infectious diseases, the concentration of MBL-A has increased approximately twice.
Applications
Mouse MBL-A ELISA kit is used for in vitro quantitative determination of mouse MBL-A in serum, plasma and cell culture supernatant.
Principle
The mouse MBL-A ELISA kit is a ready-to-use solid-phase enzyme-linked immunosorbent assay based on the sandwich principle. The working time is 3 hours. The effective form of the plate with twelve disposable 8-well plates allows free choice of batch size for determination. Samples and standards are captured by specific antibodies bound to the solid phase. The biotinylated tracer antibody will bind to the captured mouse MBL-A. The streptavidin-peroxidase conjugate will bind the biotinylated tracer antibody. The streptavidin-peroxidase conjugate will react with the substrate tetramethylbenzidine (TMB). The enzyme reaction is terminated by adding citric acid. Measure the absorbance at 450nm with a spectrophotometer. Draw a standard curve by plotting the absorbance (linear) relative to the corresponding concentration (logarithm) of the mouse MBL-A standard. The concentration of the mouse MBL-A sample running simultaneously with the standard can be determined from the standard curve.
Size
1 x 96 det.
Detection Method
Enzyme immunoassay (ELISA) technique
Detection Limit
The measurable concentration range is 1.2-75 ng/ml.
Sensitivity
The minimum concentration which can be measured is 1.2 ng/ml mouse MBL-A.
Sample
Serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids
Specimen Collection
Blood samples were collected using sterile venipuncture techniques, and serum was obtained using standard procedures. At least 5 ml of whole blood is recommended. Let the blood clot in the serum test tube for 60-65 minutes at room temperature (20-25°C). Centrifuge the blood sample and transfer the cell-free serum to a clean test tube. The serum must be handled correctly to prevent complement activation in vitro. Serum should be frozen in a sealed tube at -70°C or lower for long-term storage or transport on dry ice. The sample should not be frozen and thawed more than once. Do not use jaundice, lipemia and hemolytic serum. You cannot use heat-inactivated serum. Plasma cannot be used.
Sample Volume
100 µl/well
Species Reactivity
Mouse
Protocol Length
2 hours
Storage
Product should be stored at 4 °C. Under recommended storage conditions, product is stable for at least six months.
