Description
Antibody-dependent cell-mediated phagocytosis (ADCP) is an important mechanism of action (MOA) of therapeutic antibodies. In vivo, ADCP can be mediated by monocytes, macrophages, neutrophils and dendritic cells through FcyRIIa, FcyRI and FcyRIIIa. Although all three receptors can participate in ADCP, it is believed that FcγRIIa is the main Fcγ receptor involved in this process.

The FcγRIIa-HADCP Reporter bioassay is a biologically based MOA-based assay that can be used to measure the potency and stability of antibodies and other biological agents that specifically bind and activate FcγRIIa. The assay consists of Jurkat cells stably expressing human FcγRIIa-H (high affinity H131 variant) and NFAT-induced luciferase.

Assay Principle

The Fc effector reporter bioassay uses an alternative reading based on the traditional primary cell-based assay: FcγR and NFAT-mediated activation of luciferase activity in effector cells. The primary cells were replaced by Jurkat cell lines stably expressing human FcγRIIaH-131 and NFAT-induced luciferase. After binding to the Fc region of the relevant antibody bound to the target cell, ADCP bioassay effector cells expressing FcγRIIa-H131 have transduced intracellular signals, which can easily quantify the NFAT-RE-mediated luciferase activity.
