Description
Three different activation pathways of complement are fused with the formation of C5 invertase, and it is the cleavage of C5 by these invertases that initiates the cleavage or terminal pathway. In contrast to activation pathways that require enzymatic cleavage, the terminal pathway relies on binding-induced conformational changes. The binding of C6 promotes the binding of C7, which changes the conformation of the complex. After C8 is bound, a variable number of C9 molecules bind to the C5b678 complex (the so-called terminal complement complex (TCC) or membrane attack complex (MAC)). The formation of TCC membrane form can cause cell lysis, or trigger a variety of cellular metabolic pathways, leading to the synthesis and release of inflammatory mediators. TCC contains neoantigens that are not present in a single natural component. Neoantigens exist in both membrane-bound (MAC) and liquid phase (SC5b-9) complexes. TCC is present in normal human plasma, <1 AU/ml, and increases in patients with complement activation.
Applications
The human terminal complement complex (TCC) ELISA kit is used for in vitro quantitative determination of human TCC in plasma, urine, cerebrospinal fluid, synovial fluid and cell culture supernatant samples.
Principle
Human TCC ELISA kit is a ready-to-use solid-phase enzyme-linked immunosorbent assay based on the sandwich principle. The working time is 3 hours. The effective format of two plates with twelve disposable 8-well strips allows free choice of batch size for determination. Samples and standards are captured by specific antibodies bound to the solid phase. The biotinylated tracer antibody will bind to the captured TCC. The streptavidin-peroxidase conjugate will bind the biotinylated tracer antibody. The streptavidin-peroxidase conjugate will react with the substrate tetramethylbenzidine (TMB). The enzyme reaction is stopped by adding oxalic acid. Measure the absorbance at 450nm with a spectrophotometer. Draw a standard curve by plotting the absorbance (linear) relative to the corresponding concentration (logarithm) of the TCC standard. The TCC concentration of the sample run at the same time as the standard can be determined from the standard curve.

Specimen Collection
Blood samples were collected using sterile venipuncture techniques, and serum was obtained using standard procedures. At least 5 ml of whole blood is recommended. Let the blood clot in the serum test tube for 60-65 minutes at room temperature (20-25°C). Centrifuge the blood sample and transfer the cell-free serum to a clean test tube. The serum must be handled correctly to prevent complement activation in vitro. Serum should be frozen in a sealed tube at -70°C or lower for long-term storage or transport on dry ice. The sample should not be frozen and thawed more than once. Do not use jaundice, lipemia and hemolytic serum. You cannot use heat-inactivated serum. Plasma cannot be used.
