Description
The monoclonal antibody MM.1 recognizes CD46, also known as membrane cofactor protein. The immunohistological investigation of rat tissues showed that CD46 is only expressed in the testis and epididymis of adult rats, more specifically, only in sperm and late sperm precursors. In contrast, in humans, CD46 is expressed in all cells and tissues except red blood cells. CD46 is used to down-regulate complement activation on host tissues. It acts as a cofactor by binding to C3b and C4b. Plasma serine protease factor I allows binding, thereby degrading C3b and C4b, and protects host cells from autologous attack. In humans, it can also serve as a receptor for measles virus. Two isoforms of CD46 are dominant and are produced by alternative splicing of a single CD46 gene. The length of the same type is different. CD46 cDNA encodes a signal sequence followed by four complement control protein domains (also called short consensus repeats (SCR)).

For immunohistology, immunofluorescence and Western blotting, dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50.
