Mannose-binding lectin (MBL), also known as mannose or mannan-binding protein (MBP), is a member of the collectins group. MBL is an important pattern recognition receptor in the innate immune system. This protein mediates innate immunity Response. For example, activation of the complement lectin pathway and phagocytosis help fight infection. MBL is an oligolectin that recognizes carbohydrates on pathogens as mannose and N-acetylglucosamine. MBL contains rich Cysteine, similar to the recognition domain of collagen and carbohydrates. The activation of MBL leads to the activation of MBL-related serine proteases (MASP's). The way activated MASP-2 cleaves C4 and C2 is the same as that of C1s through the classical pathway (CP) Similarly, leading to the formation of C4b2a, the classical pathway cleaves invertase C3, and finally activates complement until the membrane attack complex is formed. MBL can activate the complement pathway without being affected by the classical complement activation pathway.
MBL is mainly synthesized by hepatocytes and has been isolated from the liver or serum of several vertebrates. Only one form of human MBL has been characterized, while two forms have been found in rhesus monkeys, rabbits, rats and mice. The mouse format is called MBL-A and MBL-C.
Compared with MBL-A, the concentration of MBL-C in serum is about 6 times. In casein and LPS injection models, MBL-A (but not MBL-C) was found to be an acute phase protein. MBL-C exists in a higher oligomeric form than MBL-A. The monoclonal antibody 14D12 is a calcium-dependent antibody.
