Description
Monoclonal antibody IIIH3 recognizes rat surfactant protein D (SP-D). SP-D belongs to the collectin family. These proteins are oligomeric proteins composed of carbohydrate recognition domains (CRD) attached to the collagen region. Although they use a different CRD structure, they are similar in structure to ficolins: the C-type lectin domain of collectin. The antimicrobial effector mechanisms of SP-D are direct conditioning, neutralization and agglutination. Therefore, infection is limited and the subsequent adaptive immune response is coordinated at the same time. The lung is the main site of SP-D synthesis, where molecules are produced by type II alveolar cells and unbranched bronchial epithelial cells and secreted to the epithelial surface. SP-D was also found in different epithelial cells of the gastrointestinal tract and epithelial cells of exocrine glands. After inflammatory stress, the synthesis and secretion of SP-D increase significantly. In inflamed lungs, increased SP-D content in lavage fluid and tissues was found, especially in type II lung cells, Clara cells and hyperplastic goblet cells. The localization of SP-D in intracellular vesicles and lysosomal particles of alveolar macrophages indicates that receptor-mediated uptake has occurred. SP-D binds to apoptotic neutrophils and enhances its clearance rate through alveolar macrophages. The monoclonal antibody IIIH3 specific to rat surfactant D has obvious cross-reactivity with human SP-D.
