Cellular Reactive Oxygen Species (ROS) Assay Kit (CAT#: CB-P048-K) Datasheet

Description
The ROS detection and analysis kit is a highly sensitive and safe detection method for detecting reactive oxygen species. ROS includes many reactive molecules and free radicals derived from molecular oxygen, which damage DNA and RNA and oxidize proteins and lipids (lipid peroxidation). The most common ROS include superoxide anion (O2-), hydrogen peroxide (H2O2), hydroxyl radical (HO-) and singlet oxygen (1O2), all of which are more reactive than oxygen (O2) itself. Molecules are produced during the electron transport process of aerobic respiration in the mitochondria, or through oxidation-reduction enzymes and metal-catalyzed oxidation.

The ROS detection and analysis kit uses the cell permeation reagent Dichlorodihydrofluorescein-diacetate (H2DCFDA), which is a fluorescent dye that can measure the intracellular hydroxyl, hydrogen peroxide and other reactive oxygen species (ROS) activity. After diffusion into the cell, the acetyl group on H2DCFDA is cleaved by intracellular esterase to generate a non-fluorescent compound, which is rapidly oxidized by ROS to highly fluorescent 2',7'-Dichlorodihydrofluorescein. The fluorescence intensity is directly proportional to the level of ROS in the cytosol.
