Description
A fusion protein consisting of the Fv portion of a monoclonal antibody linked to the 38 kDa fragment of Pseudomonas exotoxin A (amino acids 365-380 deleted). The LMB-2 immunotoxin targets the interleukin 2 receptor (also known as IL-2R or CD25), which is expressed on activated normal T cells and B cells, macrophages, and cells of various hematological malignancies.

Antibody Description
The antibody recognizes the alpha subunit of the interleukin 2 (IL-2) receptor and blocks the interaction of the cytokine with its growth factor receptor. The scientific basis for selecting the IL-2 receptor alpha subunit as a monoclonal antibody-mediated therapeutic target for leukemia / lymphoma is that few normal cells express IL-2Rα, and in patients with multiple leukemias / lymphomas Abnormal T-cell lymphomas express this receptor. In 1997, the antibody was approved by the FDA to prevent renal transplant rejection. In addition, anti-Tac provides an effective treatment for certain patients with T-cell malignancies and a series of inflammatory autoimmune diseases. Finally, treatment with this antibody with the 90Y antibody has led to a clinical response in most adult T-cell leukemia patients. These insights about the IL-2 / IL-2 receptor system have promoted the development of effective daclizumab antibody therapy for certain leukemia / lymphoma patients.

PE38 is a 38 kDa truncated form of Pseudomonas exotoxin (PE) containing amino acids 253–364 and 381–613. Pseudomonas exotoxin (or exotoxin A) is an exotoxin produced by Pseudomonas aeruginosa. Vibrio cholerae produces a similar protein called Cholix toxin (Q5EK40). It inhibits elongation factor 2. This is achieved by ADP ribosylation of EF2 using NAD +. This then causes the extension of the polypeptide to stop. This mechanism is similar to diphtheria toxin.

The antibody attaches to IL-2R on the cell membrane, promoting the entry of exotoxin. Exotoxin induces caspase-mediated apoptosis of tumor cells in part through mechanisms involving mitochondrial damage. It also catalyzes the transfer of ADP ribose from nicotinamide adenine dinucleotide (NAD) to elongation factor 2 in eukaryotic cells, thereby inactivating elongation factor 2 and inhibiting protein synthesis.
