Description
A recombinant immunotoxin, consisting of anti-CD25 monoclonal antibody RFT5 fused to deglycosylated ricin A chain (dgA), has potential anti-tumor activity. The monoclonal antibody portion of the RFT5-dgA immunotoxin is attached to CD25 (the α chain of the IL-2 receptor complex) on the cell membrane; after internalization, the dgA portion cleaves 4324 ribose and adenine bases in 28S ribosomal RNA The N-glycosidic bond between them leads to inactivation of ribosomes, inhibition of protein synthesis and cell death. CD25 is expressed on activated normal T cells and B cells and macrophages, and is frequently upregulated in many hematological malignancies.

Deglycosylated ricin A chain (dgA) is an N-glycoside hydrolase composed of 267 amino acids. Ricin is a toxalbumin, a biological toxin whose mechanism of action is inhibition of protein synthesis (manufacture of proteins) in eukaryotic cells; cell death results from the absence of proteins.
