Cathepsin G (CTSG, EC 3.4.21.20) is a serine protease that exists in the eosinophilic granules of neutrophil polymorphonuclear leukocytes. The encoded protease has a specificity similar to that of chymotrypsin C, and may be involved in the killing and digestion of phagocytic pathogens, as well as in the remodeling of connective tissue at sites of inflammation. Creative Biolabs' cathepsin G inhibitor screening kit utilizes the ability of active cathepsin G to cleave pNA (4-nitroaniline)-based synthetic peptide substrates to release pNA, which can be easily quantified using a colorimetric microplate reader. In the presence of cathepsin G inhibitors, the cleavage of this substrate is reduced/eliminated, resulting in reduced or complete loss of pNA absorbance. This simple and high-throughput adaptability assay kit can be used to screen/research/characterize potential inhibitors of cathepsin G.
