Annexin V-Cy3 Apoptosis Kit Plus (CAT#: CB-P030-K) Datasheet

This assay is based on the observation that after initiating cell apoptosis, cells quickly transfer membrane phosphatidylserine (PS) from the inner surface of the plasma membrane to the cell surface. Once it reaches the cell surface, PS can be easily detected by staining with the fluorescent conjugate of Annexin V, which is a protein with a strong natural affinity for PS. The one-step dyeing process only takes 10 minutes. In addition, the assay can be performed directly on live cells without fixation.

Annexin V-Cy3 Cell Apoptosis Detection Kit Plus includes Annexin V-Cy3, SYTOX green dye and binding buffer. SYTOX green dye does not penetrate living cells and apoptotic cells, but makes necrotic cells have strong green fluorescence by binding cell nucleic acid. After staining the cell population with Annexin V-Cy3 and SYTOX green dye in the provided binding buffer, apoptotic cells show red fluorescence, dead cells show green fluorescence, and live cells show little or no fluorescence. These populations are easily distinguished by fluorescence microscopy or flow cytometry using FITC and Rhodamine filters, where SYTOX Green dye uses FL1 channel (eg 488 nm / Em. 530 nm) and Annexin V-Cy3 uses FL2 channel (for example, 543 nm) /Em.570 nm).
