Description
Total carbohydrate assay is a simple, sensitive and reliable for detecting almost all carbohydrates.
The carbohydrate assay protocol is based on phenol-sulfuric acid assay. In this assay, polysaccharides (mono, di, third, etc.) and their derivatives are hydrolyzed into monomers in the presence of sulfuric acid and converted into furfural or hydroxyfurfural. They react with the developer to form chromogens, which can be quantified by measuring the absorbance at λ = 490 nm.
The total carbohydrate assay kit can detect most forms of carbohydrates, including simple and complex carbohydrates, glycans, glycoproteins and glycolipids.

Total carbohydrate assay protocol summary:
- add samples and glucose standards to wells
- add concentrated sulfuric acid and incubate for 15 min at 90°C
- add developer and incubate for 5 min
- analyze with a microplate reader
