SGN-10 is composed of translocation and ADP-ribosylation domain encoding Pseudomonas exotoxin (PE40) and variable heavy chain (V (H)) and variable light chain (V (L) of BR96 monoclonal antibody region. The antibody is specific for Lewis (Y) (Le (Y))-associated carbohydrate antigens expressed on various cancers.

PE40, carboxyl terminus 40 kDa of PE without its binding domain. Pseudomonas exotoxin (or exotoxin A) is an exotoxin produced by Pseudomonas aeruginosa. Vibrio cholerae produces a similar protein called Cholix toxin (Q5EK40). It inhibits elongation factor 2. This is achieved by ADP ribosylation of EF2 using NAD +. This then causes the extension of the polypeptide to stop. This mechanism is similar to diphtheria toxin.

The antibody portion mediated selective binding to target cells, while the toxin portion mediated translocation into the cytosol and subsequent cell killing.
