gC1qR exists in plasma and extracellular matrix. This molecule is an abnormally acidic single-chain protein with an apparent molecular weight of 33 kDa. It does not have conventional sequence motifs compatible with the transmembrane segment, nor does it have a common site for GPI anchors. gC1qR migrates as a single chain under both reducing and non-reducing conditions, but under non-dissociation conditions, it appears as an oligomer in gel filtration. Its multimer formation may be the key process for the gC1qR molecule to increase its affinity for multivalent ligands (such as C1q). It has been shown that gC1qR inhibits complement activation by preventing the binding of C1q to antibodies, which indicates that the binding site of gC1qR and the binding site of immune complexes existing in the spherical "head" of C1q may be located at the same position. gC1qR can interact with several proteins related to coagulation, namely thrombin, prothrombin, heparin-bound form of vitronectin, ternary complex, vitronectin-thrombin-antithrombin, and high molecular weight kinin Original and Hageman factors. In addition to its role in the complement pathway, gC1qR also participates in the enhancement of Fc receptor and CR1-mediated phagocytosis, the procoagulant activity of platelets, and the effect on mast cells, eosinophils, neutrophils and fibroblasts. Chemotactic activity. gC1qR is expressed in a variety of cells and can be used as a binding site for plasma and microbial proteins. Its antigenic sites may be recessive in suspension cells, but will be exposed when the cells are activated or fixed by the bifunctional cross-linking agent. Perhaps it is also expressed on the cell membrane as a trimer. Cross-linking or activation can therefore lead to the assembly of the gClqR trimer, and subsequent conformational changes within the molecule, exposing the originally hidden epitope. A form of gC1qR was also found in the cell. It has been shown that intracellular gC1qR binds to the cytoplasmic tail of α1B-adrenergic receptor and binds to PKCμ.
Applications
The human gC1qR ELISA kit is used for the in vitro quantitative determination of human gC1qR in plasma samples and cell culture supernatants.
Principle
Human gC1qR ELISA kit is a ready-to-use solid-phase enzyme-linked immunosorbent assay based on the sandwich principle. The working time is 3 hours. The effective form of the plate with twelve disposable 8-well plates allows free choice of batch size for determination. The samples and standards are incubated in microtiter wells coated with antibodies that recognize human gClqR. The biotinylated tracer antibody will bind to the captured human gClqR. The streptavidin-peroxidase conjugate will bind the biotinylated tracer antibody. The streptavidin-peroxidase conjugate will react with the substrate tetramethylbenzidine (TMB). The enzyme reaction is stopped by adding oxalic acid. Measure the absorbance at 450nm with a spectrophotometer. Draw a standard curve by plotting the absorbance (linear) with respect to the corresponding concentration (logarithm) of the human gClqR standard. The concentration of the human gC1qR sample running at the same time as the standard can be determined from the standard curve.

Specimen Collection
Blood samples were collected using sterile venipuncture techniques, and serum was obtained using standard procedures. At least 5 ml of whole blood is recommended. Let the blood clot in the serum test tube for 60-65 minutes at room temperature (20-25°C). Centrifuge the blood sample and transfer the cell-free serum to a clean test tube. The serum must be handled correctly to prevent complement activation in vitro. Serum should be frozen in a sealed tube at -70°C or lower for long-term storage or transport on dry ice. The sample should not be frozen and thawed more than once. Do not use jaundice, lipemia and hemolytic serum. You cannot use heat-inactivated serum. Plasma cannot be used.
