Description The recombinant immunotoxin polypeptides includes a CD3 binding domain and a Pseudomonas exotoxin variant, and particularly include a single-chain (sc) Fv as the CD3 binding moiety. scFv (UCHT-1) -PE38 is used in the treatment of transplant rejection and induction of tolerance, and treating autoimmunity and other immune disorders.

Toxic MOA PE38 is a 38 kDa truncated form of Pseudomonas exotoxin (PE) containing amino acids 253–364 and 381–613. Pseudomonas exotoxin (or exotoxin A) is an exotoxin produced by Pseudomonas aeruginosa. Vibrio cholerae produces a similar protein called Cholix toxin (Q5EK40). It inhibits elongation factor 2. This is achieved by ADP ribosylation of EF2 using NAD +. This then causes the extension of the polypeptide to stop. This mechanism is similar to diphtheria toxin.
The antibody portion mediated selective binding to target cells, while the toxin portion mediated translocation into the cytosol and subsequent cell killing.
