Description A recombinant immunotoxin, which is composed of the Fv part of anti-CD22 antibody RFB4 and the fragment of Pseudomonas exotoxin A with potential antitumor activity. The BL22 immunotoxin binds to CD22, an antigen expressed in B-cell malignancies, thereby delivering its toxin directly to tumor cells. The toxin induces caspase-mediated apoptosis of tumor cells in part through mechanisms involving mitochondrial damage. It also blocks translational extension by binding to elongation factor 2 in eukaryotic cells.

Toxic MOA PE38 is a 38 kDa truncated form of Pseudomonas exotoxin (PE) containing amino acids 253–364 and 381–613. Pseudomonas exotoxin (or exotoxin A) is an exotoxin produced by Pseudomonas aeruginosa. Vibrio cholerae produces a similar protein called Cholix toxin (Q5EK40). It inhibits elongation factor 2. This is achieved by ADP ribosylation of EF2 using NAD +. This then causes the extension of the polypeptide to stop. This mechanism is similar to diphtheria toxin.
