Cathepsin E (CTSE, EC: 3.4.23.34) is a gastric aspartyl protease that can act as a homodimer connected by disulfide bonds. This protease has a specificity similar to pepsin A and cathepsin D. It is an intracellular protease with the highest concentration on the surface of gastric epithelial mucus producing cells. It is the first aspartic protease expressed in the fetal stomach and is found in more than half of gastric cancers. Creative Biolabs' cathepsin E inhibitor screening kit utilizes the ability of cathepsin E to cleave MCA-based synthetic peptide substrates to release free MCA, which can be easily quantified using a fluorometer or fluorescent microplate reader. In the presence of cathepsin E-specific inhibitors, the cleavage of the substrate is reduced/eliminated, resulting in a reduction or complete loss of MCA Fluorometric. This simple and high-throughput adaptability assay kit can be used to screen/research/characterize potential inhibitors of cathepsin E.
