Mannose binding lectin (MBL) also known as mannose or mannan binding protein (MBP) is a member of the collectins group. MBL is an oligolectin that can recognize carbohydrates as mannose and N-acetylglucosamine on pathogens. MBL contains a recognition domain rich in cysteine, similar to collagen and carbohydrates. It forms a complex with C1r/C1, such as the serine protease MASP, which cleaves C4, C2, and C3 through proteolysis. MBL can activate the complement pathway independently of the classical and alternative complement activation pathways. The MBL-MASP pathway (more called the lectin pathway) is independent of antibodies and C1q. MBL exhibits complement-dependent antibacterial activity and directly acts as a conditioning agent, so it plays an important role in innate immunity. MBL is synthesized by hepatocytes and has been isolated from the liver or serum of several vertebrates. Only one form of human MBL has been characterized, while two forms have been found in rhesus monkeys, rabbits, rats and mice. The mouse format is called MBL-A and MBL-C. Compared with MBL-A, the concentration of MBL-C in serum is about 6 times. In casein and LPS injection models, MBL-A was found to be an acute phase protein, but not MBL-C. MBL-C exists in a higher oligomeric form than MBL-A. Monoclonal antibody 2B4 is a calcium-dependent antibody.
