Description
C5 is synthesized as a single polypeptide chain in the liver and is present in serum at a concentration of 50-80 μm/ml. Before secretion, the molecule is glycosylated and secreted into the plasma as a 190 kDa glycoprotein, which is composed of disulfide-linked α-chain (111 kDa) and β-chain (75 kDa). In functional complement tests, monoclonal antibody BB5.1 has been shown to precipitate two chains of C5 from normal mouse serum and inhibit C5-dependent hemolysis. In addition, during heart and kidney transplantation, BB5.1 administration completely inhibited the terminal complement activity of antibody-mediated rejection (AMR) in the murine model. In another mouse model, the pretreatment and intervention of monoclonal antibody BB5.1 both attenuate the disease development during glomerulonephritis induced by anti-MPO IgG. BB5.1 prevents C5 activation can prevent renal ischemia-reperfusion injury by inhibiting late-stage cell apoptosis and inflammation. In lupus disease, anti-IL-10/anti-C5 (BB5.1) combination therapy can both prevent and reduce the impact of humoral immune response.
