Single colon antibody 2/11 recognizes the activated fragment of mouse complement protein C3. The complement system is an important part of the humoral response in innate immunity and consists of three different pathways. The third complement component C3 is the core of the classical immune system. The alternative pathway of complement activation and the activation product of the complement cascade of the lectin pathway contain new epitopes that are not present in various natural ingredients.
The synthesis of C3 is tissue-specific and is regulated in response to a variety of stimulants. C3 is the most abundant protein in the complement system, and the serum protein level is about 1.3 mg/ml. Hereditary C3 deficiency makes people vulnerable to frequent attacks from bacterial infections. In ulcerative colitis and idiopathic chronic inflammatory bowel disease, the deposition of C3 in the diseased mucosa has been reported.
The proteolysis of certain enzymes causes the cleavage of C3 into C3a and C3b. C3b attaches to immune complexes and is further cleaved into iC3b, C3c, C3dg and C3f. Monoclonal antibody 2/11 is specific for cleaved C3 fragments C3b, iC3b and C3c and activated C3. Therefore, the positive reactivity in the tissue is related to the activation of the complement cascade and C3 lysis. In the case of acute inflammation, many C3 are processed into products recognized by 2/11, so they can be used as markers of inflammation. Under chronic inflammatory conditions, minimal reactivity with 2/11 can be observed. In this case, the C3dg product is mainly located in the inflammation (C3c is cleared) that the antibody 2/11 does not recognize. The long-term processing/activation of C3 is at a low level, which will reduce the detection of C3 fragments C3b, iC3b and C3c.
After detecting C3 fragments C3b, iC3b and C3c, monoclonal antibody 2/11 inhibited the hemolytic activity of mouse complement in a dose-dependent manner.

Application Notes
The diluent used depends on the detection system used. It is recommended that users test reagents and determine their own optimal dilution. The typical initial working dilution is 1:50.
