Description
The monoclonal antibody 561 recognizes the N-terminus of C5. This antibody reacts with both intact C5 (190 kDa) and C5a (115 kDa).
The complement system is composed of more than 30 proteins, which can be activated in response to tissue damage, invasion of pathogens or other foreign surfaces. The complement pathway can be divided into activation pathway and lysis pathway. The activation pathway leads to the cleavage of the fifth complement component C5 into C5a and C5b through C3, leading to activation of the lytic pathway
C5a was first described as a C5 cleavage product with chemotactic and allergic properties. Further characteristics indicate that C5a is an important part of the innate immune response, and there is now evidence that it may also play a role in adaptive immunity. Complement fragment C5a is a 74-residue pro-inflammatory polypeptide. C5a induces smooth muscle contraction, increases vascular permeability, causes degranulation of mast cells and basophils, and releases lysosomal enzymes. In addition, C5a stimulates the directed migration of neutrophils, eosinophils, basophils and monocytes. C5a binds to at least two seven transmembrane domain receptors, C5aR (C5R1, CD88) and C5L2 (gpr77). They are commonly expressed on a variety of cells, but especially in immune cells such as macrophages and mesophils The surface expression of neutrophils and T cells. The former is a recognized receptor that initiates G protein-coupled signaling through the mitogen-activated protein kinase pathway, thereby inducing the synthesis of cytokines (such as TNF-α, IL-1β, -IL-6 and IL-8) . Its blocking effect in the body greatly reduces inflammation damage. Little information is known about C5L2. The occupation of C5L2 by C5a does not trigger an increase in intracellular Ca(2+). The extensive expression of C5a receptors in the body allows C5a to trigger a wide range of effects. Application Notes
The diluent used depends on the detection system used. It is recommended that users test reagents and determine their own optimal dilution. The typical initial working dilution is 1:50. In order to neutralize the biological activity, in vitro dilution must be carried out according to the amount of C5/C5a to be inactivated.
