Monoclonal antibody 557 recognizes the epitopes of complement factor 5 (C5) and C5a. The complement system is composed of more than 30 proteins, which can be activated in response to tissue damage, pathogens or other foreign body invasion. The complement pathway can be divided into activation pathway and lysis pathway. .
The activation pathway leads to the cleavage of the fifth complement component C5 via C3. C5a was first described as a C5 cleavage product with chemotactic and allergic properties. Further characteristics indicate that C5a is an important part of the innate immune response, and there is now evidence that it may also play a role in adaptive immunity. Complement fragment C5a is a 74-residue pro-inflammatory polypeptide. C5a induces smooth muscle contraction, increases vascular permeability, causes degranulation of mast cells and basophils, and releases lysosomal enzymes. In addition, C5a stimulates the directed migration of neutrophils, eosinophils, basophils and monocytes. C5a binds to at least two seven transmembrane domain receptors, C5aR (C5R1, CD88) and C5L2 (gpr77). They are commonly expressed on a variety of cells, but especially in immune cells such as macrophages and mesophils The surface expression of neutrophils and T cells. Antibody 557 can inhibit the binding of C5a and C5a receptors through a competitive mechanism, and it will not prevent the cleavage of C5 into C5a and C5b.
