Background
In immunology, the CD3 (cluster of differentiation 3) T-cell co-receptor is a protein complex and is composed of four distinct chains. In mammals, the complex contains a CD3γ chain, a CD3δ chain, and two CD3ε chains. These chains associate with a molecule known as the T-cell receptor (TCR) and the ζ-chain to generate an activation signal in T lymphocytes. The TCR, ζ-chain, and CD3 molecules together comprise the TCR complex.

Function
Part of the TCR-CD3 complex exists on the surface of t lymphocytes and plays an important role in the adaptive immune response. When antigen-presenting cells (APCs) activate T cell receptors (TCR), TCR-mediated signals are transmitted through the cell membrane through the CD3 chain CD3D, CD3E, CD3G, and CD3Z. The cytoplasmic domains of all CD3 chains contain tyrosine-based immune receptor activation motifs (ITAMs). When TCR is involved, these motifs are phosphorylated by Src family protein tyrosine kinases LCK and FYN, leading to the activation of downstream signaling pathways. In addition to the role of signal transduction in the activation of T cells, CD3E also plays an important role in the correct development of T cells. The assembly of TCR-CD3 complex is initiated by forming two heterodimers, CD3D/CD3E and CD3G/CD3E. The endocytic sequence that exists through the cytoplasmic region of CD3E participates in the internalization of the TCR-CD3 complex and the downregulation of the cell surface.
