Function
Part of the TCR-CD3 complex exists on the surface of t lymphocytes and plays an important role in the adaptive immune response. When antigen-presenting cells (APCs) activate T cell receptors (TCR), TCR-mediated signals are transmitted through the cell membrane through the CD3 chain CD3D, CD3E, CD3G, and CD3Z. The cytoplasmic domains of all CD3 chains contain tyrosine-based immune receptor activation motifs (ITAMs). When TCR is involved, these motifs are phosphorylated by Src family protein tyrosine kinases LCK and FYN, leading to the activation of downstream signaling pathways (PubMed: 2470098). In addition to the signal transduction role of T cell activation, CD3D also plays an important role in the differentiation of thymocytes. Indeed, it is involved in the correct assembly and surface expression of the TCR-CD3 complex in the cell. If there is no functional TCR-CD3 complex, thymocytes cannot differentiate normally. It interacts with CD4 and CD8 to establish a functional link between TCR and the co-receptors CD4 and CD8, which is necessary for CD4 or CD8 t cell activation and positive selection (PubMed:12215456).
