Background
The protein encoded by this gene is a membrane glycoprotein that that mediates cell-cell signaling and plays a critical role in the growth and development of multiple organ systems. An extraordinary variety of different isoforms are produced from this gene through alternative promoter usage and splicing. These isoforms are expressed in a tissue-specific manner and differ significantly in their structure, and are classified as types I, II, III, IV, V and VI. Dysregulation of this gene has been linked to diseases such as cancer, schizophrenia, and bipolar disorder (BPD). [provided by RefSeq, Jun 2014]

Function
Direct ligand for ERBB3 and ERBB4 tyrosine kinase receptors. Simultaneous recruitment of ERBB1 and ERBB2 co-receptors leads to ligand-stimulated tyrosine phosphorylation and activation of ERBB receptors. Various isoforms have multiple functions, such as inducing the growth and differentiation of epithelial cells, glial cells, neuronal cells and skeletal muscle cells; inducing the expression of synaptic vesicle acetylcholine receptors during the formation of neuromuscular junctions and stimulating mammary lobe Bulbs sprouting and lactation, induce breast tumor cell differentiation; stimulate Schwann cell proliferation; suggest the development of myocardium, such as the developing heart trabecula. Isoform 10 may play a role in the development of motor and sensory neurons. Combined with ERBB4. Combined with ERBB3. As the ligand of integrin, and (through the EGF domain) binds to integrin ITGAV:ITGB3 or ITGA6:ITGB4. It binds to integrin and then forms a ternary complex with integrin and ERRB3, which is essential for NRG1-ERBB signal transduction. Induces the phosphorylation and activation of MAPK3/ERK1, MAPK1/ERK2 and AKT1. Ligand-dependent endocytosis of ERBB4 is essential for nrg1-mediated activation of these kinases in neurons (similarity).

Proteolytic cleavage close to the plasma membrane on the external face leads to the release of the soluble growth factor form. N- and O-glycosylated. Extensive glycosylation precedes the proteolytic cleavage (By similarity).
