Background
This gene encodes a member of the beta chemokine receptor family, which is predicted to be a seven transmembrane protein similar to G protein-coupled receptors. The gene is preferentially expressed by immature dendritic cells and memory T cells. The ligand of this receptor is macrophage inflammatory protein 3 alpha (MIP-3 alpha). This receptor has been shown to be important for B-lineage maturation and antigen-driven B-cell differentiation, and it may regulate the migration and recruitment of dentritic and T cells during inflammatory and immunological responses. Alternatively spliced transcript variants that encode the same protein have been described for this gene.

Function
C-C type chemokine CCL20 receptor. It binds to CCL20 and then transmits signals by increasing intracellular calcium ion levels. Although CCL20 is its main ligand, it can also act as a receptor for non-chemotactic ligands such as defensins. Binding of the defensin DEFB1 leads to an increase in intracellular calcium and cAMP levels. Its combination with DEFB1 is essential for DEFB1 to regulate sperm motility and bactericidal activity. Combines with defensins DEFB4 and DEFB4A/B and mediates its chemotaxis. The ligand-receptor pair CCL20-CCR6 is responsible for the chemotaxis of dendritic cells (DC), effector/memory t cells and b cells, and skin and mucosal surfaces and pathology under steady-state and inflammatory conditions, including cancer and various Plays an important role in this autoimmune disease. The signal mediated by ccr6 is essential for the immune response of intestinal mucosal microorganisms and the regulation of the inflammatory response caused by tissue damage and trauma. CCR6 is essential for recruiting pro-inflammatory IL17 to produce helper T cells (Th17) and regulatory T cells (Treg) to the site of inflammation. It is necessary for the normal migration of Th17 cells in peyers-patch and other intestinal-related tissues, and plays a role in regulating the balance and distribution of effector T cells in the inflammatory intestine. It plays an important role in coordinating the migration events of early thymocyte precursors. It is important for the subsequent normal thymocyte precursor development, but is not necessary for the formation of normal thymus natural regulatory T cells (nTregs). Optimal differentiation of DN2 and DN3 thymocyte precursors is required. It is very important for b cells to locate in the subepithelial vault of peyers-patch and for the effective isotype conversion of b cells to IgA in peyers-patch. The proper anatomical distribution of memory b cells in the spleen and the secondary recall response (through similarity) of memory b cells are essential. It positively regulates sperm activity and chemotaxis by binding to CCL20.
