Description
The recombinant anti-GPNMB immunotoxin comprises a disulfide-linked Fv (VH/VL) anti-GPNMB antibody fragment recombinantly connected to a 38 kD cytotoxic truncation of Pseudomonas exotoxin A ("PE", the 38 kD truncation is known in the art as "PE38"). PE38 is created by deleting from the 613 amino acid residue sequence of native PE residues 1-252, which correspond to domain I, along with residues 365-380 of domain Ib.

Background
The protein encoded by this gene is a type I transmembrane glycoprotein which shows homology to the pMEL17 precursor, a melanocyte-specific protein. GPNMB shows expression in the lowly metastatic human melanoma cell lines and xenografts but does not show expression in the highly metastatic cell lines. GPNMB may be involved in growth delay and reduction of metastatic potential. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

PE38 is a 38 kDa truncated form of Pseudomonas exotoxin (PE) containing amino acids 253–364 and 381–613. Pseudomonas exotoxin (or exotoxin A) is an exotoxin produced by Pseudomonas aeruginosa. Vibrio cholerae produces a similar protein called Cholix toxin (Q5EK40). It inhibits elongation factor 2. This is achieved by ADP ribosylation of EF2 using NAD +. This then causes the extension of the polypeptide to stop. This mechanism is similar to diphtheria toxin.

The antibody portion mediated selective binding to target cells, while the toxin portion mediated translocation into the cytosol and subsequent cell killing.
