Description The recombinant anti-GPC3 immunotoxin comprises a human sdAb (HN3) fragment recombinantly connected to a a wild‐type Pseudomonas exotoxin PE domain. The treatment of human sdAb (HN3) immunotoxin targeting Glypican-3 (GPC3) can lead to effective tumor regression by blocking protein synthesis and down-regulating the Wnt signaling pathway.

Background Cell surface heparan sulfate proteoglycans are composed of a membrane-associated protein core substituted with a variable number of heparan sulfate chains. Members of the glypican-related integral membrane proteoglycan family (GRIPS) contain a core protein anchored to the cytoplasmic membrane via a glycosyl phosphatidylinositol linkage. These proteins may play a role in the control of cell division and growth regulation. The protein encoded by this gene can bind to and inhibit the dipeptidyl peptidase activity of CD26, and it can induce apoptosis in certain cell types. Deletion mutations in this gene are associated with Simpson-Golabi-Behmel syndrome, also known as Simpson dysmorphia syndrome. Alternative splicing results in multiple transcript variants. Toxic MOA PE24 is a 24 kDa fragment of Pseudomonas exotoxin A. It specifically targets and kills cancer cells, and then endocytosis occurs. After proteolytic degradation, the PE28 fragment catalyzes ADP ribosylation and EF2 inactivation when introduced into the cytoplasmic sol, resulting in inhibition of protein synthesis and cell death.
The antibody portion mediated selective binding to target cells, while the toxin portion mediated translocation into the cytosol and subsequent cell killing.
