Toxic MOA Gelonin is called "ribosomal inactivation protein" and has been concerned by many researchers as a potential cancer treatment method. Gelonin is a protein toxin of approximately 30 kDa found in the seeds of the Himalayan plant Polygonum multiflorum. In a cell-free system, gelonin exerts a powerful N-glycosidase activity on the 28S rRNA unit of the eukaryotic ribosome by cleaving adenine at 4324. Gelonin lacks a carbohydrate binding domain, which exists in the form of β-chains, and therefore cannot pass through the plasma membrane, so it is very effective in cell-free systems.
The antibody portion mediated selective binding to target cells, while the toxin portion mediated translocation into the cytosol and subsequent cell killing.
