Function
Cells uptake iron into specialized endosomes through receptor-mediated endocytosis of the ligand-occupying transferrin receptor. Endosomal acidification results in the release of iron. Subsequently, the transferrin receptor complex circulates to the cell surface, the pH value returns to neutral, and the affinity of transferrin for its receptor is lost. Transferrin receptor is necessary for the development of red blood cells and nervous system. The other ligand is the immobilized hemochromatosis protein HFE, which competes with transferrin for the overlapping binding sites of the c-terminal. Positive regulation of T cell and B cell proliferation through iron absorption. (Microbial infection) As the recipient of New World Arenavirus: Guanarito, Junin and Machupo viruses.
Post-translational modifications
N- and O-glycosylated, phosphorylated and palmitoylated. The serum form is only glycosylated. Proteolytically cleaved on Arg-100 to produce the soluble serum form (sTfR). Palmitoylated on both Cys-62 and Cys-67. Cys-62 seems to be the major site of palmitoylation.
