Background
The protein encoded by this gene is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This gene is expressed most abundantly in peripheral blood leukocytes, and mediates host response to Gram-positive bacteria and yeast via stimulation of NF-kappaB.

Function
Cooperate with LY96 to mediate the innate immune response to bacterial lipoproteins and other microbial cell wall components. Cooperate with TLR1 or TLR6 to mediate the innate immune response to bacterial lipoproteins or lipopeptides. Through the action of MYD88 and TRAF6, it leads to nf-kpa-b activation, cytokine secretion and inflammation. It can also activate immune cells and promote apoptosis in response to the lipid portion of lipoproteins. Cooperating with TLR6 to recognize mycoplasma macrophage activated lipopeptide-2kd (MALP-2), soluble tuberculosis factor (STF), phenol soluble modulator (PSM) and b.b.g dorferi outer surface protein A lipoprotein (OspA-L). m.t etuberculosis PstS1 stimulates monocytes in vitro, mainly through the receptor mediated the activation of p38 MAPK and ERK1/2, but also partially through TLR4. MAPK activation against bacterial peptidoglycans also occurs through this receptor. As receptors for M. tuberculosis lipoproteins LprA, LprG, LpqH and PstS1, some lipoproteins are dependent on other co-receptors (TLR1, CD14 and/or CD36); lipoproteins act as agonists to regulate the response of antigen-presenting cells to pathogens . Mycobacterium tuberculosis HSP70 (dnaK) but not HSP65 (groEL-2) stimulates nf-kpa-b expression through this protein. Recognizes EsxA (ESAT-6), the main t antigen of Mycobacterium tuberculosis, which inhibits downstream myd88-dependent signaling (shown in mice) (similarities). According to the ligands, activation clusters composed of several receptors are formed. These clusters trigger signals on the cell surface and then target the Golgi apparatus through a lipid raft-dependent pathway. Diacylated lipopeptides form TLR2 cluster: TLR6: CD14: CD36; triacylated lipopeptides form TLR2 cluster: TLR1: CD14. The normal intake of substances necessary for Mycobacterium tuberculosis is inhibited by Mycobacterium tuberculosis LppM (due to similarity).
