Official Name
PLA2G5
Full Name
phospholipase A2, group V
Background
This gene is a member of the secretory phospholipase A2 family. It is located in a tightly-linked cluster of secretory phospholipase A2 genes on chromosome 1. The encoded enzyme catalyzes the hydrolysis of membrane phospholipids to generate lysophospholipids and free fatty acids including arachidonic acid. It preferentially hydrolyzes linoleoyl-containing phosphatidylcholine substrates. Secretion of this enzyme is thought to induce inflammatory responses in neighboring cells. Alternatively spliced transcript variants have been found, but their full-length nature has not been determined. [provided by RefSeq, Jul 2008]

Related Pathways
Its related pathways are Sweet Taste Signaling and Acyl chain remodelling of PE.
Function
Secretion of calcium-dependent phospholipase A2, which mainly acts on extracellular phospholipids. Hydrolyzes the ester bond of fatty acyl group (phospholipase A2 activity) attached to the sn-2 position of phospholipid, preferentially releasing fatty acyl groups with low degree of unsaturation, such as oleyl (C18:1) and linoleyl (C18:2) groups . Hydrolysis of low-density lipoprotein (LDL) phospholipids releases unsaturated fatty acids, which promotes the differentiation of macrophages to the M2 phenotype (through similarity). Can act in an autocrine and paracrine manner. Contribute to the lipid remodeling of the cell membrane at different subcellular locations and the production of lipid mediators to participate in pathogen clearance. Sn2 fatty acid acyl chain, the main component of lytic mitochondrial inner membrane and bacterial membrane. Promotes the phagocytosis of bacteria in macrophages by producing lysophosphatidylethanolamine. It has bactericidal activity against Gram-positive bacteria by directly hydrolyzing the phospholipids of bacterial membranes. By controlling phagosome-lysosome fusion and phagosome maturation (through similarity), phagocytosis and killing of ingested fungi are promoted. Involved in the biosynthesis of cysteinyl leukotrienes (CysLTs) in myeloid cells. In eosinophils, the release of arachidic acid and LTC4 synthesis are triggered in a pla2g4a-independent manner. In neutrophils, the cyst yslts biosynthesis initiated by PLA2G4A is expanded. By stimulating the synthesis of CysLTs to promote the clearance of macrophage immune complexes, CYSLTR1 triggers phagocytosis through CYSLTR1 (similarity). It is possible to modulate antigen processing in antigen presenting cells (through similarity). Activate Group 2 innate lymphoid cells (similarity) required for the regulation of IL33 production in lung macrophages. It may play a role in the biosynthesis of n-acylethanolamine and regulate energy metabolism. The n-acylphosphatidylethanolamine is hydrolyzed to n-acyl lysophosphatidylethanolamine, which is further cleaved by lysophospholipase D to release n-acylethanolamine.

This enzyme lacks one of the seven disulfide bonds found in similar PA2 proteins.
