Description
Monoclonal antibody RDIII-7 recognizes complement acceleration promoting factor (DAF), also known as CD55. RDIII-7 recognizes the common extracellular SCR region of CD55 and detects all isotypes of the protein. Cells express several types on their surface. The protein complement attack of defense proteins, namely C receptor I (CR1), decay accelerating factor (DAF), membrane cofactor protein (MCP) and CD59. CR1, DAF and MCP regulate complement by accelerating the decay of C3 and C5 convertase The activation pathway (CR1, DAF) or a cofactor of serine protease I, the factor cleavage irreversibly inactivates C3b (CR1, MCP).
Rat DAF (CD55) is a 60 kDa transmembrane protein that binds to C3b and C4b and can inhibit the formation and half-life of C3 convertase. It belongs to the complement activation (RCA) family of receptors. DAF is widely distributed in cells in contact with plasma complement proteins, including hematopoietic and non-hematopoietic cells. Although DAF has no important role in controlling hemolysis of red blood cells, it plays an important role in regulating the deposition of C3 on nucleated cells.
DAF works with other complement regulators to protect one's own cells from damage mediated by autologous complement. DAF and CD46 work together to avoid the deposition of autologous C3, while CD59 inhibits this pathway at a key endpoint.

Application Notes
For immunohistology, flow cytometry and Western blotting, dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50. For functional studies, in vitro dilutions have to be optimized in user's experimental setting.
