Description
The monoclonal antibody TLD-1C11 reacts with the rat complement regulator Crry. Complement contains the soluble serum protein system, which directly causes cell damage after being activated by antibodies (classical pathway), sugar (lectin pathway), or bacterial and foreign body surfaces (alternative pathway). Complement is constantly activated at low levels in serum, requiring cells to protect themselves from damage. In humans, cells are protected from complement-mediated damage by the expression of membrane-bound complement regulatory proteins that act at server points along the cascade. Human cells are protected by decay promoting factors (DAF, CD55), membrane cofactor proteins (MCP, CD46) and membrane attack complex inhibitor protein 1 (MIN1, CD59). In addition, rat analogs of DAF, MCP and CD59 have been characterized. Rats and mice express a unique activation pathway of membrane complement modulators, called type 1 complement receptor-related gene Y (Crry). In rats, there are two isoforms of Crry, with six or seven short consensus repeats (SCR), encoding 65-70 and 75-85 kDa proteins, respectively, which are anchored across the membrane. Although Crry has the functional characteristics of DAF and MCP, it cannot replace them. All rat blood cells express Crry, while approximately 37% and 39% of rat T cells express DAF and CD59, respectively. Studies in rats have shown that despite the lack of DAF and CD59, these double-negative rat T cells are more sensitive to homologous complement lysis than cells expressing DAF and CD59. However, only blocking Crry, another known rat complement regulator, can only enhance lysis on double negative cells. In rats, Crry exerts a powerful control role in the activation pathway, where it acts both as a cofactor and as a decay promoter that inactivates invertase. Crry has been successfully expressed and refolded in bacteria. The refolded protein has completely complementary regulatory activities in vitro and has been used in rat models of arthritis and other inflammatory diseases. The monoclonal antibody TLD-1C11 can be used to block the autoimmune phenomenon of rats.
