Function
Serine/threonine protein kinase is involved in different processes, such as membrane blistering and apoptotic body formation in the DNA damage response and MAPK cascade activated by MAPK14/p38 MAPK stress. Phosphorylate itself, MBP, activate MAPK8, MAP2K3, MAP2K6 and tubulin. Through the specific activation and phosphorylation of upstream MAP2K3 and MAP2K6 kinases, the MAPK14/p38 MAPK signaling pathway is activated. For DNA damage, the MAPK cascade activated by p38/MAPK14 stress may be involved in the G2/M transitional DNA damage checkpoint by mediating the phosphorylation of upstream MAP2K3 and MAP2K6 kinases. Isoform 1, but not Isoform 2, plays a role in apoptotic morphological changes, including cell contraction, membrane blistering, and apoptotic body formation. This function requires the activation of MAPK8/JNK and the nuclear localization of the c-terminal truncated isoform 1, which may be related to the death pathway related to mitochondrial casp9. Isoform 1 binds to microtubules and affects the organization and stability of microtubules independently of kinase activity. It can inhibit the activation of CHUK mediated by map3k7, thereby inhibiting the activation of nf-kpa-b, but not the activation of MAPK8/JNK. May play a role in the osmotic stress-mapk8 pathway. Heterotype 2, not Heterotype 1, is required for PCDH8 endocytosis. After homophilic interaction between the extracellular domains of PCDH8, isoform 2 phosphorylates and activates MAPK14/p38 MAPK, which in turn phosphorylates isoform 2. This process leads to the co-internalization of PCDH8 and CDH2. Both of these subtypes are involved in the phosphorylation of MAPK14.
