Background
The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a core motif of eight cysteines, seven of which are found in this factor. This gene product only forms homodimers and, therefore, does not dimerize with the other three family members. It differs from alpha and beta members of this family in having an unusual N-terminal domain, the CUB domain. Two splice variants have been identified for this gene. [provided by RefSeq, Jul 2008]

Function
Growth factors that play an important role in embryonic development, cell proliferation, cell migration, survival and chemotaxis. A potent mitogen for cells of mesenchymal origin. Play an important role in wound healing. Induces recruitment of macrophages, increases interstitial pressure, and vascular maturation during angiogenesis. It can trigger events leading to mesangial proliferative glomerulonephritis, including the influx of monocytes and macrophages and the production of extracellular matrix (similar).
Post-translational modifications
Activated by proteolytic cleavage. Proteolytic removal of the N-terminal CUB domain releasing the core domain is necessary for unmasking the receptor-binding epitopes of the core domain. Cleavage after Arg-247 or Arg-249 by urokinase plasminogen activator gives rise to the active form.
