Background
This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.

Function
Serine protease inhibitor. Inhibit TMPRSS7. It is the main inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU). As a PLAT inhibitor, it is necessary for down-regulation of fibrinolytic protein and is responsible for controlling the degradation of thrombus. As a PLAU inhibitor, it participates in the regulation of cell adhesion and proliferation. As a regulator of cell migration, it is independent of protease inhibitors. It is necessary to stimulate the migration of keratinocytes during the repair of skin damage. It is involved in cell and replication senescence. Plays a role in aging of lung alveolar type 2 cells (similar). Participate in the regulation of the cementoblast differentiation of periodontal ligament stem cells, and regulate the dentin differentiation and dentin formation in the process of dentin formation.

Description
The antibody molecules which bind plasminogen activator inhibitor 1 (PAI-1) are useful for treating diseases and conditions caused or exacerbated by fibrosis, such as systemic lupus erythromatosus (SLE), scleroderma, pulmonary fibrosis (wherein the disease is not idiopathic pulmonary fibrosis (IPF)), diabetic nephropathy, lupus nephritis, graft versus host disease, glomerulonephritis, focal segmental glomerulosclerosis, membranous nephropathy, mesangial proliferative glomerulonephritis, membranoproliferative glomerulonephritis, renal fibrosis, and COPD.
