Full Name Macrophage Stimulating 1 Receptor

Background This gene encodes a cell surface receptor for macrophage-stimulating protein (MSP) with tyrosine kinase activity. The mature form of this protein is a heterodimer of disulfide-linked alpha and beta subunits, generated by proteolytic cleavage of a single-chain precursor. The beta subunit undergoes tyrosine phosphorylation upon stimulation by MSP. This protein is expressed on the ciliated epithelia of the mucociliary transport apparatus of the lung, and together with MSP, thought to be involved in host defense. Alternative splicing generates multiple transcript variants encoding different isoforms that may undergo similar proteolytic processing. [provided by RefSeq, Jan 2016]

Function Receptor tyrosine kinases conduct signals from the extracellular matrix to the cytoplasm by binding to MST1 ligands. Regulate many physiological processes, including cell survival, migration, and differentiation. Ligand binding induces autophosphorylation of RON in its intracellular region on the cell surface, providing a docking site for downstream signal molecules. After the ligand is activated, it interacts with the pi3-kinase subunit PIK3R1, PLCG1 or adaptor GAB1. The recruitment of these downstream effectors by RON leads to the activation of multiple signal cascades, including RAS-ERK, PI3 kinase-akt or PLCgamma-PKC. RON signal activates the wound healing response by promoting epithelial cell migration, proliferation and survival of the wound site. By regulating the migration and phagocytic activity of macrophages, it plays a role in the innate immune response.
