Background This gene encodes a member of a small family of secreted growth factors that binds heparin and responds to retinoic acid. The encoded protein promotes cell growth, migration, and angiogenesis, in particular during tumorigenesis. This gene has been targeted as a therapeutic for a variety of different disorders. Alternatively spliced transcript variants encoding multiple isoforms have been observed.

Function It secretes proteins that act on cytokines and growth factors, and mediates signals through cell surface proteoglycan and non-proteoglycan receptors. It binds to cell surface proteoglycan receptors through its chondroitin sulfate (CS) group. Thereby regulating many processes such as inflammation, cell proliferation, cell adhesion, cell growth, cell survival, tissue regeneration, cell differentiation and cell migration. Participate in the inflammatory process by exerting two different activities. First, through the cooperation of LRP1 and ITGB2 and the direct effect of inducing the expression of chemokines, neutrophils and macrophages are recruited to the inflammation site. This inflammation can be accompanied by epithelial cell survival and smooth muscle cell migration after kidney and vascular injury, respectively. Second, inhibit the development of nephrogenic dendritic cells, thereby inhibiting the differentiation of regulatory T cells, and promote T cell expansion through NFAT signaling and Th1 cell differentiation. It interacts with heparin sulfate proteoglycan (similar) to promote the survival and growth of neural precursor cells and play a role in the brain. Combined with PTPRZ1, it promotes neuron migration and embryonic neuron survival. Combined with SDC3 or GPC2, it mediates neurite growth and cell adhesion. Combining chondroitin sulfate E and heparin results in inhibition of neuronal cell adhesion induced by GPC2 binding. Binds CSPG5 and promotes the elongation of oligodendrocyte precursor cell-like cells (by similarity). Also binds ITGA6:ITGB1 complex; this interaction mediates mdk-induced neurite outgrowth. In the process of arteriogenesis, by inducing the expression and release of VEGFA, and then inducing the expression of nitric oxide synthase, it plays a role in the proliferation of vascular endothelial cells. In addition, vasodilation is activated by nitric oxide synthase activation (similarity). By inhibiting the Wnt/ -catenin signal in osteoblasts (through similarity), it negatively regulates the response of mechanical load to bone formation. In addition, it also plays a role in hippocampus development, working memory, auditory response, early fetal adrenal development and female reproductive system (similar).
