Background
Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMPs are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. The enzyme encoded by this gene degrades type IV and V collagens. Studies in rhesus monkeys suggest that the enzyme is involved in IL-8-induced mobilization of hematopoietic progenitor cells from bone marrow, and murine studies suggest a role in tumor-associated tissue remodeling.

Function

Cleavage type IV and V collagen into c-terminal 3/4 large fragments and shorter n-terminal 1/4 fragments. Degrades fibronectin, but does not degrade laminin or pz peptide.

Description
GS-5745 is a monoclonal antibody that inhibits matrix metalloproteinase 9 (MMP9), an extracellular enzyme involved in matrix remodeling, tumor growth, and metastasis. Inhibiting MMP9 is expected to block paracrine signaling and metastasis and to alter the immune environment within the tumor. Preliminary safety data demonstrate a manageable safety profile for GS-5745 alone and in combination with chemotherapy. The study has been expanded to enroll additional subjects with pancreatic and esophagogastric cancer to identify potential pharmacodynamic and predictive biomarkers of response.
